immunomodulator raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
Reviewed 2026-06-28. Anything still debated is marked as such rather than presented as settled.
Overall evidence quality varies considerably. A large share of published reports come from single centers, rely on surrogate immunological markers, or lack adequate control groups. Systematic reviews have highlighted this heterogeneity as a barrier to pooling results. Open questions include which patients, if any, might benefit, what treatment duration is appropriate, and whether any effect is independent of standard care. The peptide is often described as an immune modulator rather than a therapy for one disease, which complicates confirmatory trial design.
Thymosin alpha 1 was identified in 1977 as a component of thymosin fraction 5, a heterogeneous preparation used in early studies of thymic function. Investigators purified the active material and determined its amino acid sequence, which enabled chemical synthesis. Work in the following decades concentrated on T-cell maturation and immune reconstitution in animals and small human cohorts. Early preparations varied in composition, so results from that period are difficult to compare with studies using defined synthetic peptide.
Clinical research has examined the peptide in chronic hepatitis B and C, as a vaccine adjuvant, and in sepsis and oncology settings. Findings across trials are mixed; some report changes in selected immune markers, while others find no clear clinical benefit. Many studies are small and define outcomes differently, which limits comparison. Regulatory approval is confined to a few countries, and the compound is not an approved drug in the United States or most of Europe.
The molecule consists of 28 amino acid residues with an acetyl group attached to the N-terminal serine. Its sequence is acidic overall, with several glutamic and aspartic acid residues distributed along the chain and no cysteine, so disulfide bridges do not form. The peptide carries a net negative charge at physiological pH. Because the N-terminus is blocked, the intact molecule resists degradation by many aminopeptidases, which contributes to its stability in biological fluids.
The peptide is generated in cells by cleavage of prothymosin alpha, a larger acidic protein encoded by the PTMA gene. Prothymosin alpha is expressed in many tissues, not only in the thymus, and its functions include nuclear roles in chromatin-related processes. The 28-residue fragment corresponds to the N-terminal portion of that precursor. How the cleavage occurs and how the fragment's concentration is regulated remain open questions; circulating amounts are small and difficult to measure reliably with routine assays.

Most published studies on thymosin alpha-1 report changes in immune measurements rather than clinical outcomes, and findings differ across designs and populations. Whether the peptide signals through one defined receptor or through several less specific interactions remains an open question. Its reported circulation half-life of a few hours complicates comparison of dosing schedules across trials. Mechanistic claims are frequently drawn from isolated cell cultures, and how far those results extend to whole organisms is unresolved.
Thymosin alpha-1 is a synthetic peptide of 28 amino acids whose sequence matches the amino-terminal region of prothymosin alpha. The chain is acetylated at its first residue and contains one disulfide bridge between two cysteine residues, which folds the molecule into a compact loop. Its molecular formula, C129H215N33O55, corresponds to a monoisotopic mass of roughly 3,106 daltons. Material used in laboratories is made by solid-phase synthesis rather than isolated from animal tissue.
Thymosin alpha 1 is approved as a medicine in several countries, including Italy and China, for indications such as chronic hepatitis B and as an immune adjuvant. It is not approved by the United States Food and Drug Administration as a therapeutic product. In research settings the peptide appears in studies of sepsis, vaccine response, and oncology support, often with mixed or inconclusive results. The evidence base is uneven, and reviews note that many trials were small. Regulatory status therefore differs widely between jurisdictions.
Thymosin alpha 1 is a synthetic 28-amino-acid peptide first isolated in 1966 from thymosin fraction 5, a bovine thymus extract. Its chain begins with an acetylated serine residue and ends with asparagine. The native peptide carries a molecular mass near 3,108 daltons. Researchers classify it as an immunomodulatory agent rather than a hormone with a single endocrine target. Early work framed it as a thymus-derived factor that supports T-cell maturation. The synthetic form used in research and clinical products matches the natural sequence.
==== Sodium ==== Cortisol promotes sodium absorption through the small intestine of mammals. Sodium depletion, however, does not affect cortisol levels so cortisol cannot be used to regulate serum sodium. Cortisol's original purpose may have been sodium transport. This hypothesis is supported by the fact that freshwater fish use cortisol to stimulate sodium inward, while saltwater fish have a cortisol-based system for expelling excess sodium.
Since the PTD, the hydroxyapatite tablets are not preferentially orientated; they are under less compressive residual stress, causing the microtubules to act as crack initiation sites. This manifests as cross-hatched shear microcracks forming at the microtubules in compression and as ring-shaped microcracks in tension. The tip of a larger crack creates a stress concentration that helps initiate microcracks around the microtubules ahead of it, consuming energy and resisting further damage. The imperfect linking of the microcrack to a larger crack also induces 'uncracked ligaments', which help arrest the larger crack. In comparison, enamel does not display the same fracture resistance, and fractures traveling across the DEJ are usually stopped within ~10 μm. The combination of the residual stress and the perpendicular orientation of the ITD mineralized collagen fibers significantly increases the fracture toughness and fatigue endurance limit along the microtubule direction.
Kidney involvement, in scleroderma, is considered a poor prognostic factor and frequently a cause of death. The most important clinical complication of scleroderma involving the kidney is scleroderma renal crisis (SRC), the symptoms of which are malignant hypertension (high blood pressure with evidence of acute organ damage), hyperreninemia (high renin levels), azotemia (kidney failure with accumulation of waste products in the blood), and microangiopathic hemolytic anemia (destruction of red blood cells). Apart from the high blood pressure, hematuria (blood in the urine) and proteinuria (protein loss in the urine) may be indicative of SRC. In the past, SRC was almost uniformly fatal. While outcomes have improved significantly with the use of ACE inhibitors, the prognosis is often guarded, as a significant number of patients are refractory to treatment and develop kidney failure. About 7–9% of all diffuse cutaneous scleroderma patients develop renal crisis at some point in the course of their disease. Patients who have rapid skin involvement have the highest risk of renal complications. It is most common in diffuse cutaneous scleroderma, and is often associated with antibodies against RNA polymerase (in 59% of cases). Many proceed to dialysis, although this can be stopped within three years in about a third of cases. Higher age and (paradoxically) a lower blood pressure at presentation make dialysis more likely to be needed. Treatments for SRC include ACE inhibitors.
It was soon found that plutonium was being excreted at a very slow rate, accumulating in test subjects involved in early human experimentation. This led to severe health consequences for the patients involved. From April 10, 1945, to July 18, 1947, eighteen people were injected with plutonium as part of the Manhattan Project. Doses administered ranged from 0.095 to 5.9 microcuries (μCi). Albert Stevens, after a (mistaken) terminal cancer diagnosis which seemed to include many organs, was injected in 1945 with plutonium without his informed consent. He was referred to as patient CAL-1 and the plutonium consisted of 3.5 μCi 238Pu and 0.046 μCi 239Pu, giving him an initial body burden of 3.546 μCi (131 kBq) total activity. The fact that he had the highly radioactive plutonium-238 (produced in the 60-inch cyclotron at the Crocker Laboratory by deuteron bombardment of natural uranium) contributed heavily to his long-term dose. Had all of the plutonium given to Stevens been the long-lived 239Pu as used in similar experiments of the time, Stevens's lifetime dose would have been significantly smaller. The short half-life of 87.7 years of 238Pu means that a large amount of it decayed during its time inside his body, especially when compared to the 24,100 year half-life of 239Pu. After his initial "cancer" surgery removed many non-cancerous "tumors", Stevens survived for about 20 years after his experimental dose of plutonium before succumbing to heart disease; he had received the highest known accumulated radiation dose of any human patient.

== Related odiferous compounds == Stench compounds have unpleasant odors. Some are used as odorizer or an odorant usually have an intense odor, which may be pleasant or not. They are sometimes used to confer a detectable odor to an odorless substance, like propane, natural gas, or hydrogen, as a safety measure.
=== Parasitic CK1s hijack mammalian CK1 pathways === Increasing evidence suggests that CK1 can be associated with infectious diseases by the manipulation of the CK1-related signaling pathways of the host cell by intracellular parasites, exporting their CK1 into the host cell. For Leishmania and Plasmodium, excreted CK1 contributes to reprogramming of the respective host cells. Possessing host functions parasitic CK1s are able to replace mammalian CK1s, thereby ensuring similar functions. Parasitic CK1s display a high level of identity towards human CK1δ TV1, suggesting that this human paralogue might be the preferred target for parasitic hijacking. The protein organization of parasitic CK1s is very similar to that of human CK1δ. All residues involved in ATP binding, the gatekeeper residue, as well as the DFG, KHD, and SIN motifs are generally conserved in parasitic CK1 sequences. This finding suggests, that they are crucial for CK1 function. However, the functions of these kinases in the parasites and more importantly their functions in the host cell are mainly unknown and remain to be investigated. CK1s from Plasmodium and Leishmania are most studied:
=== Taiwan === In October 2006, Taiwan banned ractopamine along with other beta-adrenergic agonists. In a 2012 climb-down, its legislature passed amendments to its Act Governing Food Sanitation, authorising government agencies to set safety standards for ractopamine. The Taiwanese Department of Health ultimately established an MRL of 10 ppb for ractopamine in beef on 31 July 2012. The American Institute in Taiwan, which represents US interests in Taiwan, states that these "and many other countries have determined that meat from animals fed ractopamine is safe for human consumption"; this is in the context of an ongoing trade dispute between Taiwan and the US on this subject, which threatened to prevent Taiwan's entry to the Trans-Pacific Partnership trade pact. In 2020, restrictions on imports of US pork with ractopamine were relaxed, leading to protests. A referendum took place on December 18, 2021, deciding whether to ban the import of pork with ractopamine, and the ban was rejected.
=== 2 June === Ukrainian officials said that during the morning two waves of air attacks were aimed at Kyiv. The Ukrainian military said they shot down 15 Russian cruise missiles and 21 drones in and around the capital overnight, with two people injured by falling debris. Five private homes were also damage according to Ukrainian officials. Another missile attack later was reported later in the day, with the Ukrainian military saying that they had intercepted all 15 cruise missiles and 18 drones launched by Russian forces and about 30 other "hostile objects". Two people were killed and four others were injured by Russian bombing in Kivsharivka, Kharkiv Oblast, while the same numbers were reported in a Russian attack on a village in Zaporizhzhia Oblast. Officials of the Russian-installed Donetsk People's Republic said that three people were reportedly killed and four others wounded by Ukrainian shelling in Makiivka. Officials in occupied Zaporizhzhia Oblast claimed that Ukrainian artillery struck a hospital camp and wounded nine people in a separate incident in Berdiansk. A Russian sympathizer and mayoral candidate in Russian-organized local elections was killed by a car bomb in Mykhailivka. The Russian Defence Ministry announced the deployment of Chechen "Akhmat" special forces to the frontlines in Mariinka, Donetsk Oblast for an offensive. Yevgeny Prigozhin said that 99% of his Wagner fighters had pulled out from Bakhmut ahead of its formal turnover to the Russian military on 5 June.

Trials differ in patient population, dose schedule, background treatment, and the endpoints used to judge success. Many are small and single-center, so random variation can dominate the reported effects.
Authorization is limited to a small number of countries and covers specific indications such as chronic hepatitis B and vaccine adjuvant use. Availability and labelling differ by jurisdiction.
Most reviews describe it as an immunomodulatory agent with an uncertain clinical effect. They generally call for larger, better-controlled trials before firm conclusions are drawn.
Thymosin fraction 5 is a mixture of many peptides obtained from thymus tissue, while thymosin alpha 1 is a single defined 28-residue molecule. The two names appear together in older literature because the purified peptide was first obtained from that mixture.
